| General description【一般描述】 | GenElute 無內毒素質粒制備試劑盒可以簡單、快速、低成本的純化質粒DNA(內毒素水平≤0.1 EU/μg)，所得產物可用于高效轉染。內毒素(也稱為脂多糖或LPS)常常會與質粒DNA共純化，這會顯著降低內毒素敏感細胞系的轉染效率。 通過離心分離法從過夜培養的物中收集重組大腸桿菌，并采用SDS改性堿裂解法處理以形成澄清的裂解液。通過簡單的萃取相分離步驟去除掉內毒素。在高鹽環境下，質粒DNA吸附到硅膠膜上接受進一步的純化。通過簡單的離心洗滌步驟處理后，在無內毒素的水中洗脫結合所得的質粒DNA。從25~200 mL培養物可以制備出多達1.2 mg不含內毒素的質粒DNA。 回收的質粒DNA主要以超螺旋形式存在。此類DNA可直接用于轉染、限制性酶切、克隆、測序和PCR擴增等下游應用。 |
